  1. 6. Calcium dynamics and vesicle-release proteins in a prion-infected neuronal cell line

    Author : Malin Sandberg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Prion; Scrapie; Voltage-gated N-type calcium channels; SNARE proteins; Exocytosis; Long-term depolarization;

    Abstract : The prion particle, PrPSc, is an infectious, misfolded form of the cellular prion protein, PrPC that is expressed at high levels in the central nervous system. Prions cause neurodegenerative diseases with clinical signs including dementia, ataxia and myoclonus. These diseases cause characteristic electroencephalographic changes. READ MORE

  2. 7. Antibody-based Profiling of Expression Patterns using Cell and Tissue Microarrays

    Author : Sara Strömberg; Fredrik Ponten; Joakim Lundeberg; Anna Asplund; Stephen Hewitt; Uppsala universitet; []
    Keywords : Pathology; antibody-based proteomics; tissue microarray; cell line; immunohistochemistry; melanocytes; image analysis; biomarker; Patologi;

    Abstract : In this thesis, methods to study gene and protein expression in cells and tissues were developed and utilized in combination with protein-specific antibodies, with the overall objective to attain greater understanding of protein function.To analyze protein expression in in vitro cultured cell lines, a cell microarray (CMA) was developed, facilitating antibody-based protein profiling of cell lines using immunohistochemistry (IHC). 3. 8. Triggering and mechanisms of natural killer cell mediated cytotoxicity

    Author : Yenan Bryceson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Natural killer cells; antibody dependent cellular cytotoxicity; natural cytotoxicity; immunodeficiency; hemophagocytic lymphohistiocytosis;

    Abstract : Natural killer (NK) cells are innate immune cells that contribute to defense against infected and transformed cells by target cell killing and cytokine release. In addition, data suggest that NK cells contribute to immune homeostasis and reproduction. READ MORE

  4. 9. Genetic and cellular studies of familial hemophagocytic lymphohistiocytosis

    Author : Eva Rudd; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Familial hemophagocytic lymphohistiocytosis; PRF1; STX11; UNC13D; degranulation; genotype; phenotype;

    Abstract : Familial hemophagocytic lymphohistiocytosis (FHL) is a rare autosomal recessive and genetically heterogeneous disorder of immune dysregulation with an incidence of 1/50000 live births that is inevitably fatal without appropriate treatment. The disease is characterized by fever, hepatosplenomegaly, cytopenias, hyperferritinemia, hypertriglyceridemia, hypofibrinogenemia and, sometimes, hemophagocytosis in bone marrow and/or other organs such as liver, spleen or lymph nodes. 5. 10. Prognostic Factors in Malignant Melanoma

    Author : Åsa Bolander; Michael Bergqvist; Simon Ekman; Anders Larsson; Daniel Brattström; Gunnar Wagenius; Torgny Rasmuson; Uppsala universitet; []
    Keywords : Malignant melanoma; prognostic factors; survival; chemotherapy; radiation;

    Abstract : Because of the failure so far to find effective treatment for patients with advanced stages of melanoma, increasing efforts have been made to find prognostic factors identifying patients in the risk zone for development of metastasis.This thesis investigates the prognostic powers of a few selected serological and immunohistochemical biomarkers.
