  1. 1. Cyclic peptides as biological tools : Applications for diagnosis and therapeutics in rheumatoid arthritis and COVID-19

    Author : Camilla Eriksson; Ulf Göransson; Sunithi Gunasekera; Christian Becker; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Scaffold peptides; peptide grafting; rheumatoid arthritis; SARS-CoV-2; autoantibodies; peptide synthesis; pharmacognosy;

    Abstract : The overall aims of the projects included in this thesis were to design, synthesize and evaluate cyclic peptide scaffolds grafted with biological epitopes for their ability to bind antibodies in rheumatoid arthritis and COVID-19 sera. One of the main outcomes of this thesis was the observation that scaffold peptides grafted with target epitopes efficiently neutralized anti-citrullinated protein autoantibodies (ACPA) from rheumatoid arthritis patients. 2. 2. Photoswitchable Peptidomimetics : Synthesis and Photomodulation of Functional Peptides

    Author : Miranda Varedian; Adolf Gogoll; Morten Grøtli; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; β-hairpin; peptidomimetics; stilbene; thioaurone; photochemistry; Organic chemistry; Organisk kemi;

    Abstract : The secondary structure of peptides is of pivotal importance for their biological function. The introduction of photoswitchable moieties into the backbones of peptides provides a unique way of regulating their conformation using an external stimulus, i.e., light. READ MORE

  3. 3. Discovery of Small Peptides and Peptidomimetics Targeting the Substance P 1-7 Binding Site : Focus on Design, Synthesis, Structure-Activity Relationships and Drug-Like Properties

    Author : Rebecca Fransson; Anja Sandström; John S Svendsen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; substance P 1-7; peptidomimetics; structure-activity relationship; drug-like properties; phenylalanine; imidazole; MAP aryl amides; carbonylation; Pharmaceutical chemistry; Farmaceutisk kemi; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Biologically active peptides are important for many physiological functions in the human body and therefore serve as interesting starting points in drug discovery processes. In this work the neuropeptide substance P 1–7 (SP1–7, H-Arg-Pro-Lys-Pro-Gln-Gln-Phe-OH), which has been demonstrated to reduce neuropathic pain and attenuate opioid withdrawal symptoms in animal models, has been addressed in a medicinal chemistry program with the overall aim of transforming this bioactive peptide into more drug-like compounds. 4. 4. Cyclotides evolve : Studies on their natural distribution, structural diversity, and activity

    Author : Sungkyu Park; Göransson Ulf; Lars Bohlin; Mohamed Marahiel; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; cyclotide; cyclic cystine knot; evolution; peptide synthesis; chemical ligation; QSAR; Viola; Violaceae; phylogeny; Farmakognosi; Pharmacognosy;

    Abstract : The cyclotides are a family of naturally occurring peptides characterized by cyclic cystine knot (CCK) structural motif, which comprises a cyclic head-to-tail backbone featuring six conserved cysteine residues that form three disulfide bonds. This unique structural motif makes cyclotides exceptionally resistant to chemical, thermal and enzymatic degradation. READ MORE

  5. 5. Chimeric gene delivery vectors : Design, synthesis, and mechanisms from transcriptomics analysis

    Author : Moataz Dowaidar; Ülo Langel; Sandrine Sagan; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Cell-penetrating peptides; magnetic nanoparticles; graphene oxide; autophagy; siRNA; SCO; QSAR; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Abstract : Delivery of nucleic acid is a promising approach for genetic diseases/disorders. However, gene therapy using oligonucleotides (ONs) suffers from low transfection efficacy due to negative charges, weak cellular permeability, and enzymatic degradation. READ MORE
