1. Design, Synthesis, Mechanistic Rationalization and Application of Asymmetric Transition-Metal Catalysts
Abstract : This thesis describes mechanistic studies, rational ligand design, and synthesis of asymmetric transition metal catalysts. The topics addressed concerned [Papers I-VII]:[I] The asymmetric addition of diethyl zinc to N-(diphenylphosphinoyl)benzalimine catalyzed by bicyclic 2-azanorbornyl-3-methanols was studied. READ MORE

2. Consequences of CYP2D6 polymorphism for the disposition and dynamics of tolterodine : a novel drug in the treatment of urinary bladder overactivity
Abstract : The pharmacokinetics and pharmacological effects of tolterodine were studied in man following administration of increasing oral and intravenous single-doses. The influence of metabolic phenotype in extensive and poor metabolizers of debrisoquine was determined. READ MORE

3. Metabolic drug interactions in man : methodological aspects on in vivo studies
Abstract : The aim of this thesis was to investigate methodological aspects on in vivo metabolic drug interaction studies in man. Five pharmacokinetic studies in healthy persons or patients are presented. Together, they represent a set of model studies to evaluate the occurrence of, and extent of metabolic drug interactions in vivo. READ MORE

4. Cytochrome P450 Enzymes in the Metabolism of Vitamin D3
Abstract : A cytochrome P450 enzyme in pig kidney that catalyzes the hydroxylations of vitamin D3 and C27-sterols was cloned. DNA sequence analysis of the cDNA revealed that the enzyme belongs to the CYP27 family. The recombinant kidney CYP27A enzyme catalyzed the 25-hydroxylation of vitamin D3 and the 27-oxygenation of C27-sterols. READ MORE

5. Development of techniques and methods for drug analysis by packed capillary liquid chromatography with octadecylbonded silica and porous graphitic carbon columns
Abstract : Packed capillary liquid chromatography in combination with ultraviolet and mass spectrometric (MS) detection was explored as an analytical tool for drug analysis. The focus was put on column and sample preparation techniques as well as on fundamental aspects and development of chromatographic separations. READ MORE
