1. Development and Evaluation of Nonparametric Mixed Effects Models
Abstract : A nonparametric population approach is now accessible to a more comprehensive network of modelers given its recent implementation into the popular NONMEM application, previously limited in scope by standard parametric approaches for the analysis of pharmacokinetic and pharmacodynamic data. The aim of this thesis was to assess the relative merits and downsides of nonparametric models in a nonlinear mixed effects framework in comparison with a set of parametric models developed in NONMEM based on real datasets and when applied to simple experimental settings, and to develop new diagnostic tools adapted to nonparametric models. 2. Novel Fatty Acid Dioxygenases of Human and Plant Pathogenic Fungi : Studies by Gene Deletion and Expression
Abstract : The dioxygenase-cytochrome P450 fusion proteins (DOX-CYP) comprise a heme-containing enzyme family that shares structural and catalytic properties with mammalian prostaglandin H (PGH) synthases. 7,8-Linoleate diol synthase (7,8-LDS) of Gaeumannomyces graminis was first characterized, and DOX-CYP enzymes are of mechanistic and biological interest. 3. Mass Spectrometric Analysis of Oxylipins : Application to Cytochrome P450-Dependent Metabolism
Abstract : Cytochrome P450 (CYP) family 4 constitutes monoxygenases responsible for hydroxylation of fatty acids and other lipids. For example, CYP4F3 metabolizes leukotrienes and CYP4F8 prostaglandin H. Importantly, six of the twelve CYP4 enzymes are orphans, i.e. READ MORE

4. Finding improved drug strategies for schizophrenia : Preclinical studies on lumateperone and sodium nitroprusside
Abstract : Schizophrenia is a severe psychiatric disorder affecting approximately 20 million people worldwide. The disease consists of positive symptoms e.g. hallucinations, negative symptoms such as anhedonia, and cognitive deficits, e. READ MORE

5. Discovery of Novel Fatty Acid Dioxygenases and Cytochromes P450 : Mechanisms of Oxylipin Biosynthesis in Pathogenic Fungi
Abstract : Dioxygenase-cytochrome P450 (DOX-CYP) fusion enzymes are present in diverse human and plant pathogenic fungi. They oxygenate fatty acids to lipid mediators which have regulatory functions in fungal development and toxin production. READ MORE
